The short answer

NMN is a precursor to NAD+, a coenzyme involved in cellular energy metabolism. Oral NMN raises blood NAD markers in human trials, and a dose-finding trial testing 300, 600 and 900 mg found blood NAD rose in every group and rose further with dose, with 600 mg appearing optimal.

The form, and why it matters

β-Nicotinamide mononucleotide, the reduced β anomer, which is the form the salvage pathway uses. Purity is stated at 99.9% because the synthesis leaves a small impurity fraction, and the figure is on the label rather than in the marketing.

Typical dose

Human trials have used 250 to 900 mg a day over 8 to 12 weeks. The dose-finding trial tested 300, 600 and 900 mg, and 600 mg came out best with 900 no better. Spark sits at 500 mg in one capsule, chosen from that trial rather than from a bigger number.

What the research shows

A multicentre double-blind trial in 80 adults aged 40 to 65 gave 300, 600 or 900 mg for 60 days: blood NAD rose dose-dependently, six-minute walking distance and quality-of-life scores improved, and biological age held steady while the placebo group's crept up. A six-week trial in 48 trained runners improved ventilatory threshold dose-dependently. A 12-week trial in 108 adults over 65 found afternoon dosing beat morning on drowsiness and lower-limb function. Trials so far are short, 8 to 12 weeks, and mostly in healthy middle-aged and older adults.

Who should be careful

The human safety data is short-term. There is no long-term safety record, and NMN is not recommended in pregnancy, breastfeeding or for under-18s. Talk to a clinician if you are managing a condition or take prescription medicine.
